Anesthesia of Epinephelus marginatus with essential oil of Aloysia polystachya: an approach on blood parameters Anais da ABC (AABC)
FOGLIARINI,CARINE O.; GARLET,QUELEN I.; PARODI,THAYLISE V.; BECKER,ALEXSSANDRO G.; GARCIA,LUCIANO O.; HEINZMANN,BERTA M.; PEREIRA,ANA MARIA S.; BALDISSEROTTO,BERNARDO.
ABSTRACT This study investigated the anesthetic potential of the essential oil (EO) of Aloysia polystachya in juveniles of dusky grouper (Epinephelus marginatus). Fish were exposed to different concentrations of EO of A. polystachya to evaluate time of induction and recovery from anesthesia. In the second experiment, fish were divided into four groups: control, ethanol and 50 or 300 µL L−1 EO of A. polystachya, and each group was submitted to induction for 3.5 min and recovery for 5 or 10 min. The blood gases and glucose levels showed alterations as a function of the recovery times, but Na+ and K+ levels did not show any alteration. In conclusion, the EO from leaves of A. polystachya is an effective anesthetic for dusky grouper, because anesthesia was...

Antagonism by hemoglobin of effects induced by L-arginine in neuromuscular preparations from rats BJMBR
Ambiel,C.R.; Alves-Do-Prado,Wilson.
Nitric oxide (NO)-synthase is present in diaphragm, phrenic nerve and vascular smooth muscle. It has been shown that the NO precursor L-arginine (L-Arg) at the presynaptic level increases the amplitude of muscular contraction (AMC) and induces tetanic fade when the muscle is indirectly stimulated at low and high frequencies, respectively. However, the precursor in muscle reduces AMC and maximal tetanic fade when the preparations are stimulated directly. In the present study the importance of NO synthesized in different tissues for the L-Arg-induced neuromuscular effects was investigated. Hemoglobin (50 nM) did not produce any neuromuscular effect, but antagonized the increase in AMC and tetanic fade induced by L-Arg (9.4 mM) in rat phrenic nerve-diaphragm...

This study evaluated the biochemical changes associated to successive captures of Salminus brasiliensis reared at different stocking densities. The experiment was conducted in tanks with a capacity of 1,000 L, connected to a recirculating system with temperature control and continuous aeration. Fish were stocked at densities of 30, 150 and 300 fish m-3 and the successive catches were applied at the end of the experimental period of 80 days. The biochemical changes were evaluated at the end of the trial through the parameters: hematocrit, hemoglobin, glucose and lactate in plasma and Na+, K+ and Cl- and Ca2+ in the gills. The concentration of glucose increased with the increase of stocking density as also registered for lactate concentration, except for...

Can the hemoglobin characteristics of vesicomyid clam species influence their distribution in deep-sea sulfide-rich sediments? A case study in the Angola Basin ArchiMer
Decker, Carole; Zorn, N.; Le Bruchec, J.; Caprais, Jean-claude; Potier, N.; Leize-wagner, E.; Lallier, F. H.; Olu, Karine; Andersen, A. C..
Vesicomyids live in endosymbiosis with sulfur-oxidizing bacteria and therefore need hydrogen sulfide to survive. They can nevertheless live in a wide range of sulfide and oxygen levels and depths, which may explain the exceptional diversity of this clam family in deep-sea habitats. In the Gulf of Guinea, nine species of vesicomyid clams are known to live in cold-seep areas with pockmarks from 600 to 3200 m deep, as well as in the organic-rich sediments of the Congo deep-sea fan at 5000 m deep. Our previous study showed that two species living in a giant pockmark have different oxygen carriers, suggesting different adaptations to hypoxia. Here, we studied the hemoglobin structure and oxygen affinity in three other species, Calyptogena valdiviae, Elenaconcha...

Early exposure to chronic hypoxia induces short- and long-term regulation of hemoglobin gene expression in European sea bass (Dicentrarchus labrax) ArchiMer
Cadiz, Laura; Servili, Arianna; Quazuguel, Patrick; Madec, Lauriane; Zambonino, Jose-luis; Mazurais, David.
European sea bass (Dicentrarchus labrax) inhabits coastal waters and may be exposed to hypoxia at different life stages, requiring physiological and behavioral adaptation. In the present study, we attempted to determine whether regulation of hemoglobin (Hb) gene expression plays a role in the physiological response to chronic moderate hypoxia in whole larvae and hematopoietic tissues (head kidney and spleen) of juveniles. We also tested the hypothesis that hypoxia exposure at the larval stage could induce a long-term effect on the regulation of Hb gene expression. For this purpose, D. labrax were exposed to a non-lethal hypoxic condition (40% air saturation) at the larval stage from 28 to 50 days post-hatching (dph) and/or at the juvenile stage from 196 to...

Effect of the inoculum size on the axenic in vitro production of Steinernema carpocapsae Ciencia e Investigación Agraria
Suárez-Sánchez,Juan; Alatorre-Rosas,Raquel; Poggi-Varaldo,Héctor M; Barrera-Cortés,Josefina.
The effect of inoculum size on the axenic in vitro production of Steinernema carpocapsae, using a transparent medium with hemoglobin and cholesterol, was studied. Axenic cultivation was performed with fertile eggs ofS. carpocapsae (hatching of eggs: 89 ± 4%), which were obtained by lyses of female nematodes and subsequent incubation. Two important aspects of this work include the use of a completely transparent medium enriched with hemoglobin and cholesterol and the high multiplication factor of the population (MFIP) of 87, which is considered high for in vitro axenic cultures of nematodes. In vivo methods have the advantage of high MFIP, but they also have the disadvantage of the high number and complexity of the operations involved for the purposes of...

Familial congenital cyanosis caused by Hb-M Yantai (α-76 GAC → TAC, Asp → Tyr) Genet. Mol. Biol.
Sun,Yanbo; Wang,Pingyu; Li,Youjie; Jiao,Fei; Li,Zunling; Ma,Ying; Li,Wei; Xie,Shuyang.
Methemoglobin (Hb-M) is a rare hemoglobinopathy in China. We hereby report on a family living in Yantai, East China, with congenital cyanosis due to Hb-M mutation. The proband, a 65-year-old female, presented 63% oxygen saturation. Both Hb-M concentration and arterial oxygen saturation remained unchanged, even following intravenous treatment with methylene blue. There was also no change in blood-color (chocolate-brown) after adding 0.1% KCN. A fast-moving band (Hb-X) in hemolysates was found by cellulose acetate electrophoresis, the Hb-X/Hb-A ratio exceeding 10%. GT transition at 131nt of exon 2, although present in one of the α2-globin alleles, was not found in α1-globin alleles as a whole. This mutation leads to the aspartic acid to tyrosine substitution...

Growth and stress of dourado cultivated in cages at different stocking densities PAB
Braun,Neiva; Dafre,Alcir Luiz; Lima,Ronaldo Lima de; Beux,Luis Fernando; Brol,Fernanda Franciele; Nuñer,Alex Pires de Oliveira.
The objective of this work was to evaluate the growth and the stress levels of juvenile dourado (Salminus brasiliensis) cultivated in cages. Fish stocked at densities of 15 (D15) and 30 (D30) fish per square meter were evaluated in a completely randomized design with three replicates. Fish were fed twice a day with extruded ration (42% crude protein). Density influenced only biomass and daily food intake, and glucose and lactate concentrations increased over time. D15 and D30 did not influence the growth of dourado. However, the increase of glucose and lactate levels over time indicates that cultivation in cages is a stressful condition for this species.

Increased fetal hemoglobin levels in patients infected with human immunodeficiency virus (HIV1/2) BJMBR
Poli-Neto,A.; Nonoyama,K.; Oshiro,M.; Ebner-Filho,W.; Miguita,K.; Medeiros,T.M.D.; Watanabe,C.I.; Barretto,O.C.O..
Fetal hemoglobin was measured in HIV1/2 patients under treatment with combined therapy (zidovudine and a protease inhibitor). A total of 143 patients and 103 normal individuals were investigated by the quantitative method of Betke and the semi-quantitative acid elution method of Kleihauer. In the normal person, hemoglobin F makes up less than 1% and an increase higher than 1.5% was observed in 21.4% of HIV patients by the method of Betke and in 24.8% of HIV-infected patients by the method of Kleihauer. The quantitative biochemical method of Betke showed that the populations were significantly different (two-tailed Mann-Whitney test). The reason for this hemoglobin F increase might be ascribed to the effect of zidovudine or to direct viral action on gamma...

Almeida,Sandra M.; Umeo,Suzana H.; Marcante,Rafael C.; Yokota,Meire E.; Valle,Juliana S.; Dragunski,Douglas C.; Colauto,Nelson B.; Linde,Giani A..
Pleurotus ostreatus is able to bioaccumulate several metals in its cell structures; however, there are no reports on its capacity to bioaccumulate iron. The objective of this study was to evaluate cultivation variables to increase iron bioaccumulation in P. ostreatusmycelium. A full factorial design and a central composite design were utilized to evaluate the effect of the following variables: nitrogen and carbon sources, pH and iron concentration in the solid culture medium to produce iron bioaccumulated in mycelial biomass. The maximum production of P. ostreatus mycelial biomass was obtained with yeast extract at 2.96 g of nitrogen L−1 and glucose at 28.45 g L−1. The most important variable to bioaccumulation was the iron concentration in the cultivation...

Polymorphism of human haptoglobin and its clinical importance Genet. Mol. Biol.
Wobeto,Vânia Peretti de Albuquerque; Zaccariotto,Tânia Regina; Sonati,Maria de Fátima.
Haptoglobin (Hp) is a plasma glycoprotein, the main biological function of which is to bind free hemoglobin (Hb) and prevent the loss of iron and subsequent kidney damage following intravascular hemolysis. Haptoglobin is also a positive acute-phase protein with immunomodulatory properties. In humans, the HP locus is polymorphic, with two codominant alleles (HP1 and HP2) that yield three distinct genotypes/phenotypes (Hp1-1, Hp2-1 and Hp2-2). The corresponding proteins have structural and functional differences that may influence the susceptibility and/or outcome in several diseases. This article summarizes the available data on the structure and functions of Hp and the possible effects of Hp polymorphism in a number of important human disorders.

Prevalence of common α-thalassemia determinants in south Brazil: importance for the diagnosis of microcytic anemia Genet. Mol. Biol.
Wagner,Sandrine C.; Castro,Simone M. de; Gonzalez,Tatiana P.; Santin,Ana P.; Filippon,Leticia; Zaleski,Carina F.; Azevedo,Laura A.; Amorin,Bruna; Callegari-Jacques,Sidia M.; Hutz,Mara H..
Alpha thalassemia has not been systematically investigated in Brazil. In this study, 493 unrelated individuals from the southernmost Brazilian state of Rio Grande do Sul were screened for deletional forms of α-thalassemia. One hundred and one individuals had microcytic anemia (MCV < 80 fL) and a normal hemoglobin pattern (Hb A2 < 3.5% and Hb F < 1%). The subjects were screened for -α3.7,-α4.2,-α20.5, -SEA and -MED deletions but only the -α3.7 allele was detected. The -α3.7 allele frequency in Brazilians of European and African ancestry was 0.02 and 0.12, respectively, whereas in individuals with microcytosis the frequency was 0.20. The prevalence of α-thalassemia was significantly higher in individuals with microcytosis than in healthy individuals...

The secondary alcohol and aglycone metabolites of doxorubicin alter metabolism of human erythrocytes BJMBR
Misiti,F.; Giardina,B.; Mordente,A.; Clementi,M.E..
Anthracyclines, a class of antitumor drugs widely used for the treatment of solid and hematological malignancies, cause a cumulative dose-dependent cardiac toxicity whose biochemical basis is unclear. Recent studies of the role of the metabolites of anthracyclines, i.e., the alcohol metabolite doxorubicinol and aglycone metabolites, have suggested new hypotheses about the mechanisms of anthracycline cardiotoxicity. In the present study, human red blood cells were used as a cell model. Exposure (1 h at 37ºC) of intact human red blood cells to doxorubicinol (40 µM) and to aglycone derivatives of doxorubicin (40 µM) induced, compared with untreated red cells: i) a ~2-fold stimulation of the pentose phosphate pathway (PPP) and ii) a marked inhibition of the...
